❶〔生涯〕a lifetime; one's whole life

一生に一度の好機だ
This is 「the chance of a lifetime [a once-in-a-lifetime chance].／This is the opportunity of a lifetime.

Such a chance never comes twice in a lifetime.

One's span of life is short.
